I don't think it matters, most carbs do not have a hole in the throttle plate (it may have been added to help balancing somehow) - what matters is setting the plates in the correct orientation and position so that they seal well with the body, and identically for both side as much as possible, to enable a smooth idle.

Throttle plates are marked with angle, plate set with this in normal orientation.
Hole will be at the bottom opposite the progression holes.
