Timing chain adjuster screw

My engine has recently returned from rebuild by a specialist. The timing chain whines, I've removed the cam cover and the chain is much tighter than the recommended 1/2" of play. I've tried to adjust, but when I slacken the adjuster screw the locknut seizes on the thread before it seats on the abutment - I can't see whether the adjuster screw thread is damaged or covered in thread lock. Now the question - can I unscrew and remove the adjuster screw to examine the thread without anything falling off the other end or becoming displaced?

The adjustment screw is removed as a unit, you need to undo the large nut this will take the adjustment screw with it, self-explanatory when you see it, however there is a coil spring and tension plunger left in the hole just leave in place it shouldn’t go anywhere.
